Partial Purification of ß-Glucanase from Fruiting Bodies of Straw Mushroom (Volvariella volvacea) by Aqueous Two Phase Extraction and Its Hydrolysis of Mushrooms Cell Walls

چکیده

The objective of this study was to partially purify ß-glucanase from crude enzyme extract of straw mushroom (Volvariella volvacea). The crude enzyme extract was prepared from the fruiting bodies of straw mushroom. The ßglucanase was partially purified using an aqueous two-phase extraction with Triton X-114 and followed by ammonium sulfate precipitation in the range of 50-80% saturation. The ß-glucanase activity was increased 4.1-fold with recovery yield of 70%. The phenolic compound was also reduced to 7.2% of the original content, avoiding the post purification tanning of the enzyme. Furthermore, the ß-glucanase was able to hydrolyze V. volvacea cell-wall and other mushrooms cell-walls such as oyster mushroom (Pleurotus sajor-carjo) and ear mushroom (Auricularia auricular).

Purification and characterization of phytase with a wide pH adaptation from common edible mushroom Volvariella volvacea (Straw mushroom).

A novel phytase with a molecular mass of 14 kDa was isolated from fresh fruiting bodies of the common edible mushroom Volvariella volvacea (Straw mushroom). The isolation procedure involved successive chromatography on DEAE-cellulose, CM-cellulose, Affi-gel blue gel, Q-Sepharose and Superdex-75. The enzyme was a monomeric protein and was unadsorbed on DEAE-cellulose, CM-cellulose and Affi-gel b...

Identification of Ergosterol in Mushrooms

Mushroom has gained popularity as prized table delicacy with high nutritive and medicinal value. Ergosterol is a biological precursor (a provitamin) to vitamin D2. The contents of vitamin D2 and sterols in some wild and cultivated mushrooms were determined. Vitamin D2 was determined using an HPLC method, including saponification and semipreparative normalphase HPLC purification Lv et al. [1]. C...

Induction of laccase activity in the edible straw mushroom, Volvariella volvacea.

Volvariella volvacea, strain V14, produces multiple forms of extracellular laccase when grown in submerged culture in a defined medium with glucose as sole carbon source, and on cotton waste 'compost' representative of the conditions used for industrial-scale mushroom cultivation. In liquid culture, enzyme synthesis is associated with the onset of secondary growth, and is positively regulated b...

Composition and Expression of Genes Encoding Carbohydrate-Active Enzymes in the Straw-Degrading Mushroom Volvariella volvacea

Volvariella volvacea is one of a few commercial cultivated mushrooms mainly using straw as carbon source. In this study, the genome of V. volcacea was sequenced and assembled. A total of 285 genes encoding carbohydrate-active enzymes (CAZymes) in V. volvacea were identified and annotated. Among 15 fungi with sequenced genomes, V. volvacea ranks seventh in the number of genes encoding CAZymes. I...

Lead Accumulation in the Straw Mushroom, Volvariella volvacea, from Lead Contaminated Rice Straw and Stubble

Straw mushrooms were grown on lead contaminated rice straw and stubble. Study materials were dried, acid digested, and analyzed for lead using flame atomic absorption spectrophotometry. The results showed the highest lead concentration in substrate was 445.350 mg kg⁻¹ in Treatment 3 (T3) and the lowest was BD (below detection) in Treatment 1 (T1). The maximum lead content in straw mushrooms was...
